ummmmmmmmmmmmmmmmmmmmmmmmAheh heh! Wow.. really got late didn’t it..!!! Haha, yeah.. I was supposed to write here every 3 days? Is that what it says? Wow! Haha! Wow. Haha.

Actually, I’m not even supposed to be writing this right now. Basically this means that I’ll only update my blog when I’m absolutely not supposed to. So that’s pretty much all of the time. I’ve been feeling very apathetic recently. Anyways, I… I can’t remember anything that happened, anything significant anyways. I’ll just talk about polypeptide synthesis because that’s what I’m supposed to be doing right now anyways.

Once upon a time, there lived a small, native hometown in the world of Cell called Nucleus. Nucleus was a cozy, charming place to be, with its own customs, culture and citizens (why am i using so many words starting with c) But Nucleus had a problem. The economy was collapsing. There was very little tourism. Nobody from outside Nucleus found this little town interesting enough, or they just didn’t know much about it at all..! Without these tourists that we will call Proteins, the nucleus, and eventually the whole Cell, and eventually the whole ORGANISM, will collapse… Nobody knew it, but Nucleus was more important than it let on. And in turn, Proteins were what kept the world churning. So one day, the wise elder in Nucleus called DNA was set to fix this problem. DNA is like.. the towns grandma which you go to for medicinal tinctures and black magic. She’s kind of the boss. DNA has decided that there needs to be a spokesperson that can spread the culture that DNA holds. DNA needs a spokesperson, because DNA is old and rickety and can’t travel outside the nucleus like all the youngins can, and even if DNA could, well.. She doesn’t speak the same language as outsiders from the nucleus do. And another problem.. She’s pretty infertile, so she can’t just make a copy of herself like that. DNA needs some help from her trusted henchman called RNA polymerase. RNA polymerase (I was going to shorten the name but RNA polymerase is really badass so I’m keeping it) is the secret miracle worker that is extremely underappreciated and doesn’t even brag about working for DNA. Nobody really knows anything about RNA polymerase. You don’t question it. I don’t question it. I’m just a bio student and this is due in 2 days. Anyways, RNA polymerase understands what must be done, and will perform open heart surgery on DNA very carefully. DNA isn’t just gonna let RNA polymerase start the surgery all willy nilly, oh no no no, because there needs to be grace to this. In the genes of DNA exists a promoter sequence, which basically tells RNA polymerase to start the process over here. RNA polymerase finds the promoter sequence and will then begin to unzip both strands of DNA away from each other, kind of prying them apart to wedge itself onto the template strand of the DNA and begin pre-mRNA synthesis!!!!!!!!!!!!!!!!!! Now, RNA polymerase is pretty smart. Nobody knows its backstory or its family or anything about it, but it somehow has knowledge of the language outside of the Nucleus. This is really important for creating the spokesperson. So okay, first let’s get nucleotides out of the way. You know how each DNA helix has these little lines through them? Those lines are called nucleotides and they are the basic building blocks for the information in the genes. If you’re a homestuck fan, you will recognise that the four main bases are C,G,A,T. In RNA, it has the same bases except the T (thymine) is changed to U (uracil) because RNA is spicy like that. However, they are both pretty much the same thing and uracil actually helps the message become translated from DNA language to RNA language. Going back to the pre-mRNA synthesis, RNA polymerase is now creating nucleotides that correspond to the ones attached to the template strand and starts knitting together a link, pushing this link out from 3’ to 5’ (basically from the inside of the dna strand to the outside). So think of this as a sort of dark web surgery, like RNA polymerase is creating a homunculus out of the cells from DNA’s armpit or something. RNA polymerase is actually pretty smart, because SINCE they are creating a link from the template strand that has its corresponding nucleotides, it’s actually making a copy of the OTHER side of the DNA strand called the non-template strand (since that strand was zipped up to the template strand before) except it’s now in RNA language. (god this is so hard to do without an image but im so not bothered to add one in) SO. Now that each nucleotide has been converted and the link has gotten preeetty long now, DNA rings the breaking bad ding ding ding thingy which tells RNA polymerase to STOP!!! STOP!!!!!!!! Yes, there is a stop signal called the terminator sequence in the dna like the promoter sequence. RNA polymerase is done…. MY MAN!!!!! YOU DID YOUR PURPOSE!!!!!! THE SURGERY WAS A SUCCESS!!!!!!!!!!!!! Politely, RNA polymerase zips up DNA back to shape and releases this beautiful creation that we talked about earlier, pre-mRNA. this baby is the rosetta stone of the RNA language. But it’s not ready yet, que the ‘pre’, as it has a long journey ahead of it before it steps out into the world to spread the message far and wide. In a prokaryotic cell (bacteria and simple celled organisms), this pre-mRNA would just be mRNA and would be ready to go, but in eukaryotic cells (humans and animals) it needs to be edited first. pre-mRNA has a lot of useless bullshit alongside the actual gems which need to be spliced out. pre-mRNA is honestly kind of weird so the aunties of the town are gonna give her a makeover before she leaves. pre-mRNA’s introns (the useless) are spliced out and the exons (gems) are condensed together to create one comprehensible young capable mRNA!!!!!!1 i simplified that process a lot but its kinda not really that important to me in the grand scheme of my life so im letting it be. NOW the pre-mRNA has turned into Mature mRNA and is… sniff is READY TO LEAVE TOWN!!!!!!!!!!!!!!!!! Did I explain it in a way you could somewhat understand, if you didn’t know about transcription before? If I did, I'd be happy.. I think everything can be easy to explain to others, no matter how hard the concept is. I think sometimes people purposefully explain things in a confusing or vague manner because they have a superiority complex and want to make people think they are dumber than they actually are. But you aren’t smarter than other people if you don’t need things explained to you.
